My family has used the self-administered oral swab at an LA City Covid testing site each time we've tested.
My son (19 yrs) tested last Friday 10/09 & received Negative result.
He was out late on Saturday & I had him retest on Monday 10/12, and he received a Positive result.
He tested again yesterday 10/16 and received a Negative result.
His father & I both tested on 10/09, 10/14, and 10/16 - all results Negative.
Very confusing. How do we proceed?

If saliva testing accuracy has been demonstrated to be equal to the nasal swab method, why subject patients to the uncomfortable nasal swab? Why is, then, all testing not saliva-based?

Have you done any tests to see how stable the samples are for longer periods of time than in this paper? Also, can you clarify which Norgen RNA purification kits you used (Cell-Free or Total RNA kits)? Thank you. Trying to get this ramped up in my lab.

Sorry for the late reply. That was the Total RNA kit. We did not see how long samples would be stable. Hope the ramp up is going well!

Thank you Noah. Can you provide more details on the specifics of your RNA extraction protocol? Given the good results you have we'd like to replicate as closely as possible and it is not fully clear what protocol you are following within the multiple iterations of the manufacturer instructions. For example, given that your samples were collected with DNA/RNA shield added, I assume you used some sort of adaptation of the "1D Lysate Preparation from Nasal or Throat Swabs"--but I can imagine multiple ways you may have adapted this. Was β-mercaptoethanol used?
